Outline of Annual Research Achievements

肺線維芽細胞においてフォークヘッド型転写因子FOXL1が比較的高く発現しており、DNA低メチル化と関連していることを見出した。さらにFOXL1はスパーエンハンサー領域とも関連していた。
肺組織におけるFOXL1の発現は免疫組織染色およびRNA in situ hybridizationで確認した。
さらにトランスクリプトーム解析により、FOXL1により制御される遺伝子群を同定した。YAP・TAZで制御される遺伝子群、PDGFα受容体、BMPリガンドやアンタゴニストの発現が、FOXL1により制御されることを見出した。
FOXL1の発現低下により、肺線維芽細胞の細胞増殖やコラーゲンゲル収縮が抑制されることも明らかになった。FOXL1は肺線維症の線維芽細胞に発現しており、その病態に関与している可能性が示唆された。
