Outline of Final Research Achievements

In the study, we investigated how to confirm the discovery of genetic mutations other than ryanodine receptors that may cause malignant hyperthermia in an experimental system. In addition to knock-in mice to observe the response of individuals to inhalational anesthetics and depolarizing muscle relaxants, it was thought that additional experiments, such as culturing myotubular cells and observing changes in intracellular calcium dynamics, should be conducted in combination.

Academic Significance and Societal Importance of the Research Achievements

悪性高熱症の原因となる遺伝子変異はそのほとんどがリアノジン受容体から見つかっているが,すべてが見つかるわけではない.今後,新たな遺伝子に存在する遺伝子変異が悪性高熱症の原因となる可能性が指摘されたとき,今回検討した実験系を使用することにより,その遺伝子変異が本当に悪性高熱症の原因となりうるのかが検証できる.新たな原因遺伝子を発見するこことは,悪性高熱症の原因を探索し治療を考えることにおいて非常に有意義なことである.
