Outline of Annual Research Achievements

骨、軟骨の欠損は骨折などの外傷や変形性関節症、感染症などにより引き起こされ、生活の質を大きく損なうにもかかわらず治療に難渋することが多い。我々は生体材料であるコラーゲンを人工合成・重合する技術を開発し、3Dプリンターで自由に造形する技術を開発した。骨・軟骨欠損部を補填する生理的な基質の作成に最適な技術と考え、作成したコラーゲンインプラントと脂肪組織由来の組織幹細胞を用いて骨軟骨欠損の新規治療法の開発に挑んた。まずI型コラーゲンゲルにヒト脂肪由来間葉系幹細胞を懸濁し、インプラントを形成した。細胞培養用容器にて培養を行い、ゲル内での増殖能、分化能、基質産生能を評価した。コラーゲン内に包埋し、通常の培地で培養した限りは、SOX9, COL2A1, COL9A1, COL11A2, Aggrecanなどの軟骨分化マーカーの劇的な増加はみられず、特徴的な分化を示すことはなかった。次に同様の実験をBMP-2、TGFbeta-1などを用いた軟骨分化誘導後のヒト脂肪由来間葉系幹細胞で行ったところ、これらの軟骨分化マーカーの良好な増加を認め、COL1A1, オステオポンチン、オステオカルシン、Runx2, Osterixなどの骨分化マーカーの増加は伴わなかった。組織切片を作成し、各種染色を実施したが、軟骨基質についても確認できた。これらの条件で作成された組織を、家兎軟骨欠損モデル(大腿骨滑車軟骨部)に移植したところ、術後8週間の時点でサフラニンO陽性の軟骨様組織が広範囲にみられた。
