Outline of Final Research Achievements

To find lncRNAs related to cancer progression and metastasis in kidney cancer, we used 2 collecting duct carcinoma samples. Finally we identified two lncRNAs which expression was significantly increased in collecting duct carcinoma tissues compared to normal kidney tissues. Two lncRNAs are GNAS and YYAIAP1, localised in cytoplasm. There two lncRNAs potentially bind to miRNA-205 and miR-371a-5p based on computer algorithms. Two miRNAs also potentially bind to MXI1 and KIF3B.
Currently their expression levels being verified in clear cell renal cell carcinoma tissues.

腎癌

Academic Significance and Societal Importance of the Research Achievements

腎癌の中での最も悪性度の高いベリニ管癌組織を使用し、癌組織で高発現しているlncRNA候補を同定後に、腎癌の中で最も多い組織系の淡明細胞癌で再検証することで、癌化、浸潤、転移に関連するlncRNAを同定できる可能性を検証した。現時点で淡明細胞癌組織内での発現解析を行っている。この実証ができれば、腎癌全体の発症、浸潤、転移に関連するマーカーとなるため、今後術後のバイオマーカーとなりうる。また、lncRNAの作用機序にmiRNAのスポンジ効果があるため、候補となるlncRNAが潜在的に結合するmiRNA,及びその標的遺伝子の解析も終了しているため、それらも淡明細胞癌組織で検証を行っている。
