Outline of Annual Research Achievements |
For stabilization and functionalization of a DNA-nanostructure, a novel macromonomer consisting of two guanidinium (Gu+) ions, a benzophenone (BP) and an anchoring azide unit was synthesized. As a proof-of-concept we first tested adhesive property of the macromonomer with a 40-base pair DNA. In gel electrophoresis, free DNA migrate towards anode but the DNA after mixing with the macromonomer did not migrate, indicating its charge neutralization. Further, the macromonomer can dimerize by irradiation with ultraviolet light and strengthens its multivalent interaction with DNA. We found that this macromonomer like-wise adhere to DNA origami nanostructures such as 6-helix bundles, cuboid and tetrahedron, and the coated DNA nanostructres showed stability against serum proteins.
