Outline of Annual Research Achievements

嗅神経には、Hyperpolarization-activated Cyclic Nucleotide-gated (HCN)チャネルの類縁であるCyclic Nucleotide-Gated(CNG)チャネルが嗅覚線毛に発現しており、cAMPは匂いのセカンドメッセンジャーとしても利用される。本研究助成を受けて我々は、嗅神経細胞に遍く存在するOlfactory Marker Protein(OMP)の中にcAMP結合部位を発見し、cAMPの結合定数を生化学実験で決定した。OMPは、cAMPの急速な吸着分子として機能し、CNGチャネルの過度の活性化とその後のNa+/Ca2+の過剰流入を防ぐ。OMPがノックアウトされた嗅神経では、頻回のcAMPの過剰負荷によりCNGチャネルによる脱分極が進むことで細胞膜が過剰順応をし、神経発火が止まる。そのためOMP-knockout(KO)マウスの嗅上皮にcAMPを負荷すると、KOマウスは目の前の物体(エサ)に興味を示すもののSniffing行動ではエサとして認識できなくなる「無嗅覚症状」を呈することを証明した(Nature Communications, 2020、BBRC, 2020)。OMPはcAMPの吸着をすることで、CNGチャネルを介した急性cAMP応答の時間分解能を高める一方、cAMPを分解から保護することでcAMPプールを安定化しHCNチャネルの持続活性化を可能にする(BBRC, 2017)。OMPのcAMP緩衝能は、一度結合したcAMPの解離を介した基底cAMPプールの形成にも重要であることが分かった(BBRC, 2020)。さらにATP、ADP, AMPといった核酸も結合することが可能であることから、OMPが細胞内核酸シャトルとして機能している可能性が示唆される(BB Report, 2021)。
