Outline of Annual Research Achievements

手根管症候群(Carpal Tunnel Syndrome, CTS)は、最も頻度の高い絞扼性神経障害として知られる。しかしながら、いまだ病因が不明であり、治療法は外科介入に限られている。我々はCTSの病態解明と新規治療法の開発として、細胞老化に着目して研究を行った。手根管症候群患者の線維化滑膜にplatelet-derived growth factor receptor alpha (PDGFRα)陽性線維芽細胞の細胞老化システムの破綻が影響するとの仮説を立てて研究を実施した。手根管症候群患者の滑膜組織からOut growth法で接着細胞を採取してPDGFRα+細胞を培養し、 p16, p14, p15など細胞老化関連因子のタンパク質発現および遺伝子発現について、フローサイトメトリー解析, 蛍光免疫染色, Real-Time-PCRなどで解析を行った。結果、手根管症候群患者の手根管滑膜において、p15を含めた細胞老化関連因子およびBcl2など抗アポトーシス関連因子の上昇を認めた。 次に、動物モデルを用いて老化細胞が手根管滑膜の線維化を誘導するか、老化細胞をモデル動物の手根管部に移植した時の手根管滑膜病態の変化を解析するため、モデル動物由来線維芽細胞を放射線照射によって細胞老化を誘導し、モデル動物の手根管部に移植した。すると、手根管滑膜の線維化が誘導されたことから、老化細胞が手根管滑膜の線維化を誘導が示唆された。次に、細胞老化除去薬として知られるdasatinib(D)、quercetin(Q)について、老化細胞への効果を検討するため、in vitroで検討を行った。それぞれ単独で使用した時、D+Qのコンビネーションで使用した時を検討し、D+Qの組み合わせで老化細胞のアポトーシスを誘導を認めた。
