Analysis of the effect of Cetuximab on fluorescence kinetics in living tongue cancer cells using the fluorescent ubiquitination-based cell cycle indicator (Fucci).

Outline of Annual Research Achievements

Fucciを導入した舌癌由来のSAS細胞を用い、Cetuximab(Cmab)の細胞周期動態に及ぼす影響を解析した。CmabをSAS-Fucci細胞に長期処理すると、細胞増殖抑制を認め、コロニー中央部にはG1 arrestした赤色蛍光を有する細胞集塊を認めた。Cdt1-mKO2の蛍光強度は、コントロール群のG1期細胞と比較し有意に高かった。またCmab処理により細胞遊走が有意に抑制された。この現象について、Western blotにてタンパクを解析した。予想に反し、EGFR経路でシグナルを受けるERK1/2のリン酸化の抑制はわずかであった。Cmab長期処理によりRBのリン酸化および非リン酸化が抑制されたため、タンパク分解が亢進している可能性が考えられた。そこで細胞周期関連タンパク群であるCDK阻害因子(CKI)についても解析したところ、Cmab長期処理後、p27の増加を認めた。p27は細胞増殖の接触阻害で機能するCKIであり、これがRBの脱リン酸化を介して細胞増殖を抑制したと考えられた。p27の分解にはSCFSkp2が関与することから、Skp2のリン酸化をみると、確かにCmab長期処理により消失を確認した。またSkp2の活性を制御するAkt・mTORの変化も解析した。その結果、Akt・mTORのリン酸化もCmab長期処理により有意に抑制された。mTORの抑制はオートファジーを誘導することも知られている。その関連因子であるAtg12とLC3Bを解析すると、Cmab長期処理でAtg12-Atg5およびLC3B-IIの有意な増加を認めた。
以上より、SAS細胞におけるCmab長期処理は、早期のEGFR経路の阻害は引き起こさないが、細胞遊走低下による、細胞接触ストレスを介したAkt・mTOR活性を低下させ、p27の増加とオートファジーの亢進により細胞増殖を抑制したと結論づけられた。
