Outline of Annual Research Achievements

PAWW stress mice were stimulated for 7 weeks. The feces were collected every week. After 7 weeks, blood, spleen, brain, Peyer's patches and Mesenteric lymph nodes were collected. IL1ra, IL1a, IL1b, IL-6 and IL-10 in lymphocyte, parts of brain, spleen, Peyer's patches and Mesenteric lymph nodes were determined by Quantitative real-time PCR. The results showed that IL6 level of PAWW stress mice was increased significantly in Peyer's patches, and Il10 level was increased significantly in Mesenteric lymph nodes. The gut microbiota from feces in PAWW stress mice were characterized by high-throughput 16S-seq. In PAWW stress mice, Proteobacteria (Xanthomonadales, Pseudomonadales, Enterobacteriales) were increased significantly, but Actinobacteria was decreased significantly at 7 weeks. These results suggested that PAWW stress mice has changing of immune system and microbiota.

Reason

I found differentiation of immune system (IL6 and IL10 level) and microbiota as we expected.

Strategy for Future Research Activity

IL6 and IL10 were selected to be first bio-marker of PAWW stress mice. New IL6 and Il10 evaluation system was developing. The goal is IL6 and IL10 evaluation system will be easier to evaluate compound screening.

Causes of Carryover

The characterization of microbiota fee was required in next fiscal year. So I did not used all money in last year.
