Outline of Annual Research Achievements

ミトコンドリアタンパク質の網羅的な理解は医薬・生物学の大きな課題となっている。特にミトコンドリアタンパク質が関与するタンパク質間相互作用 (PPI)については、創薬標的としての価値が高いもののほとんど調べられておらず、また情報も散在している状況である。本研究課題においてミトコンドリアタンパク質の網羅的なPPI=インタラクトームを解明するための技術を開発することを目的とする。PPI予測技術MEGADOCKを活用し、網羅的にPPI予測を実施してデータベースを構築し、関連情報を統合化して公開する。さらにin vitro実験による未知PPIの発見を試み、これまで見過ごされていた多くの標的を世界に先駆けて調べることができる情報基盤として本研究のデータベースが利用可能であることを示す。
今年度(2年目)は、MEGADOCKの並列計算方式およびGPU実装についての改良を行い、MEGADOCK 5.0としてソフトウェアのバージョンアップを行った。また、MEGADOCK 5.0の産総研ABCIスーパーコンピューターでの並列化効率を計測し、rt_Fノードを512基用いた場合に96.4%となる並列化効率 (strong scaling) となることを示した。また、MEGADOCKを用いたミトコンドリアタンパク質の網羅的な予測結果をMEGADOCK-Web-Mitoとして整理し、MitoProteomeデータベースに含まれるミトコンドリアタンパク質の情報を結びつけて検索可能なシステムを構築、公開した。
