How do potassium ions regulate the formation for non-canonical structures of nucleic acids and tumor progressions?

Outline of Annual Research Achievements

本研究では、がん細胞内で低下していると考えられる「カリウムイオンが核酸非二重らせん構造の形成を調節し、がんなどの疾患の発症、進行を制御している」という新しい仮説を提示し、証明することを目的とした。まず、前年度までの研究において、正常細胞、悪性度の異なるがん細胞中における細胞内環境を評価した。その結果、がんの悪性度に応じて発現するがんの増殖に関わるイオンチャネルは、カリウムイオンを細胞外に放出する役割も担うことを見出し、さらにこれらの細胞内では、G-四重らせん構造を鋳型にもつDNAから転写される転写量が変化することも見出した (J. Am. Chem. Soc., 140,642 (2018))。つまり、細胞内では、細胞内の環境変化に応じてG-四重らせん構造の安定性が変化し、転写量を制御していることを見出した。
そこで2019年度は、種々の疾患にかかわる遺伝子を対象に細胞内環境変化に応じた非二重らせん構造の構造解析および、これらの非二重らせん構造が転写や翻訳などの生体反応に変異に及ぼす影響を解析した。まず、擬似細胞内環境下において、三重らせん構造に及ぼすイオンの効果を解析した結果、通常は三重らせん構造が解離するカリウムイオン濃度でも擬似細胞内環境下では安定な三重らせん構造が形成された(Molecules, 25, 707 (2020))。さらに、四重らせん構造は、パラレル型、アンチハイブリット型などのトポロジーによって複製反応及ぼす影響が異なり(Sci. Rep., 10, 2504 (2020))、これらの四重らせん構造のトポロジーは、共存溶質の水酸基との相互作用によって制御されていることを見出した(Biochem. Biophys. Res. Commun., 525, 177 (2020))。分子動力学計算を基に設計した、疾患細胞内における環境でも四重らせん構造を安定に形成できる人工核酸の開発にも成功した(Molecules, 25, 387 (2020)、 Nucleic Acids Res, 48, 3975 (2020))。
