Outline of Final Research Achievements |
This work aims application of dynamic nuclear polarization NMR to observe water soluble biomolecules such as proteins with ultrahigh sensitivity by developing water soluble DNP-enhancing molecules. A DNP-enhancing pentacene is a extremely hydrophobic molecule with no water solubility. We developed a two water-soluble cyclodextrins-linked molecule with inner space for appropriate pentacene inclusion and hydrophilic surface. We next examined preparation of a pentacene that two cyclodextrins are covalently bound by using Click reaction.

Academic Significance and Societal Importance of the Research Achievements |
動的超偏極法(DNP)は、核磁気共鳴分光法(NMR)の欠点である測定感度の低さを大きく補う優れた手法である。ただし、DNPの促進に優れた機能を有するペンタセンは極度に疎水的であるため、測定に時間のかかるタンパク質などの水溶性分子には応用できなかった。このペンタセンを水溶化して上記目的にかなわせるために、ペンタセンの形状分子の包接に適した内腔を有する水溶性分子の合成を行った。今後この分子に適したペンタセン誘導体を包接させることで、タンパク質などの構造解析の大幅効率化に寄与することが期待される。
