In vitro mouse small cell lung cancer model using isolated lung epithelium

Outline of Annual Research Achievements	令和２年度においては、小細胞肺癌mouse model (Tp53(-); CCSPrtTA; tet0cre; floxedRb1; floxed Hes1)および( Tp53(-); CCSPrtTA; tetOCre; floxedRb1; floxedhes1; tet0 mutant EGFR)の妊娠マウス５腹より、胎仔（胎齢16日を採取した。胎仔肺より、酵素処理後に上皮を単離し、matrigelに包埋し、器官培養を行った。培養後200-300日すると、培地内に遊出する腫瘍細胞を観察することが出来た。 (Tp53(-); CCSPrtTA; tet0cre; floxedRb1; floxed Hes1)マウスより３種類、(Tp53(-); CCSPrtTA; tetOCre; floxedRb1; floxedhes1; tet0 mutant EGFR)より２種類の細胞を得ることが出来た。これら細胞について、Western blotting, 免疫染色、及び、免疫不全マウスの皮下に移植した組織を解析したところ、小細胞肺癌は、(Tp53(-); CCSPrtTA; tetOCre; floxedRb1; floxedhes1; tet0 mutant EGFR)マウスより１株のみ得ることが出来た。
