Outline of Annual Research Achievements

多孔質膜を挟んで上下二つの流路を有し、上下の流路には電極を設置して多孔質膜を介した電流の抵抗値(経上皮電気抵抗:TEER)を計測できるようにしたデバイスを作製した。そのデバイスの上流路から腸管上皮細胞(Caco2のサブラインであるC2BBe1細胞)を流し込み多孔質膜上にC2BBe1細胞を接着させ、上下に培地を流しながら、TEER値を計測および電顕観察から、C2BBe1細胞の密着結合が多孔質膜全体に形成されたことを確認した。
そこで、このデバイスの細胞側に対して上層の培地から青色蛍光タンパク質遺伝子導入大腸菌(AmCyan/Ecoli)を導入し、新鮮な培地を流し続けた。上層の流速を調節することにより大腸菌数を一定にすることに成功し、大腸菌と共にC2BBe1細胞も安定的に培養を継続できることに成功した。蛍光観察により、大腸菌はコロニーを形成してC2BBe1細胞状にとどまっていることが判明し、電顕観察でも確認ができた。また、C2BBe1細胞上へ乳酸菌L.murinusを播種し、ある流速条件下ではC2BBe1細胞が最低でも3日間に渡り、密着結合を維持、L.murinusは一定生菌数で維持し、大腸菌と同様にコロニーを形成していることが確認できた。
C2BBe1細胞上でのL.murinusとAmCyan/Ecoliの共培養を行った。L.murinusをC2BBe1細胞上に1日間培養し生着させ、その後AmCyan/Ecoliを播種し、共培養を行った。C2BBe1細胞の密着結合は1日間維持されることを確認した。また、C2BBe1細胞上にはL.murinusおよびAmCyan/Ecoliの各々のコロニーが形成されていることが確認できた。以上により、2種類の腸内細菌を腸管上皮細胞上で共培養し、細菌数を維持できる流体デバイス系の確立に成功した。
