Outline of Annual Research Achievements

睡眠中に記憶が固定・消去されていると考えられているが、どのように調節されているのか神経科学的に解明されていない。本研究では、我々が独自に睡眠覚醒調節と記憶制御の両方に重要であることを同定した視床下部のメラニン凝集ホルモン(MCH)神経を中心に解析を行う。Kilduff博士(SRI International, Director)との密接な共同研究によって、最新の光技術を駆使した神経活動の光操作と光記録をMCH神経に適用し、記憶を評価する行動実験と組み合わせることによって、睡眠時の記憶制御メカニズムについて神経回路レベル、細胞レベルにて詳細に明らかにすることを目的としている。MCH神経特異的にカルシウムインジケーターを発現せて、ファイバーフォトメトリーを用いてMCH神経活動を記録したところ、覚醒時とレム睡眠時に活動が認められた。さらに、Kilduff博士との共同研究において、超小型顕微鏡を用いて意識下に自由行動するマウスのMCH神経活動を1細胞レベルで記録した。その結果、MCH神経には、覚醒時に活動するタイプと、レム睡眠時に活動するタイプが存在することが明らかになった。そのどちらのタイプのMCH神経が記憶に関わっているのかについて明らかにするために、光遺伝学を用いた脳状態依存的なMCH神経活動抑制と記憶の評価を行った。その結果、覚醒時に抑制しても記憶に変化はなかったが、レム睡眠時に抑制したところ記憶成績が向上した。これらのことからレム睡眠時に活動するMCH神経が記憶抑制に関わっていることが明らかになった。これらの結果をまとめてScience誌に報告した(Izawa et al., Science 2019)。

Reason

MCH神経の1細胞レベルでの活動記録と脳状態依存的な光遺伝学を用いた抑制を組み合わせて、レム睡眠時に活動するMCH神経が記憶抑制に関わっていることを明らかにすることに至った。これらの結果をまとめてScience誌に報告した(Izawa et al., Science 2019)。

Strategy for Future Research Activity

MCH神経が遊離するどの伝達物質が記憶に関わっているのかについて、分子レベルでメカニズムを明らかにする。インビトロとインビボ実験を組み合わせて分子レベルでの検討を行う。

Causes of Carryover

コロナウイルスなどの影響があり、海外渡航を控えたため。コロナウイルスの影響がなくなれば、次年度に鹿児島大学の山下哲助教がSRI Internationalに長期間滞在し、実験を行う予定となっている。
