Research Abstract

H21年度は以下の成果を得た。
1.血小板活性化因子(PAF)産生酵素であるリゾPAFアセチル転移酵素を2種類(LPCAT1とLPCAT2)同定することに成功した。この内、LPCAT2がリン酸化されることによって活性調節されていることを明らかにし、そのリン酸化部位を同定した。
2.多発性硬化症モデルマウスの脊髄病変部におけるリピドミクス解析により、プロスタグランジン合成酵素mPGES-1依存的なプロスタグランジンE2産生が、Th1及びTh17型免疫応答を調節して、多発性硬化症の病態を増悪させていることを明らかにした。多発性硬化症の病変部ではmPGES-1を発現しているマクロファージから、大量にPGE2が産生されて免疫応答を調節し、結果として症状を増悪させていることが示唆された。
3.骨密度および骨形態計測学的パラメターの測定により、ロイコトリエンB_4(LTB_4)第一受容体(BLT1)ノックアウトマウスでは、卵巣摘除およびLPS投与による骨吸収が顕著に抑制されていることを見出した。破骨細胞ではLTB_4が産生され、BLT1は発現していたがBLT2は発現していなかった。また、BLT1ノックアウトマウス由来破骨細胞のカルシウム吸収能は、野生型マウス由来破骨細胞に比較し減少していた。破骨細胞の形態変化と破骨細胞活性には密接な関係があることを考慮すると、LTB_4は、パラクライン・オートクライン的に作用して破骨細胞活性を増加することが示唆された。
4.GPCRの膜貫通領域(TM)内にはGPCR間で保存されたアミノ酸残基が幾つか存在するが、これら残基に変異を有するPAF受容体(PAFR)は小胞体内貯留すること、そして、特異的リガンドを薬理学的シャペロンとして作用させることでこの貯留PAFRが細胞膜へと移送されることを明らかにした。ところで、作製した変異体の内、TM6内のPro^<247>変異PAFRは膜表面に移行させたものをアゴニスト刺激した場合、その後の細胞内移送過程にも異常を有することがわかった。
