Research Abstract

高等動物において嗅覚は、求餌や生殖行動、天敵や腐敗物からの忌避など、個体や種の生存にとって不可欠な情報受容システムである。マウスの場合、多様な匂い情報を約1000種類のodorant receptor (OR)で識別するからくりは、嗅球における匂い情報の二次元展開にあるとされている。即ち、嗅細胞の軸索が、発現するORの種類に従って嗅球上の特定の糸球に投射するため、嗅上皮で受容された匂い分子の結合情報は、嗅球表面ではどの糸球が発火したかという位置情報に変換される。
本研究課題では、ORの種類によって指令的に生じる嗅細胞の軸索投射について、嗅細胞の神経個性を構成する軸索誘導分子や接着・反発分子が、どの様な転写制御を受けているのかの解明を目指した。嗅球の前後軸に沿った軸索投射ついては、それぞれのOR分子に固有なレベル生み出されるcAMPのシグナルの強度で転写量が決まる反発分子、NrplとSema3Aを介した軸索間相互作用によって制御されていることを示した。一方背腹軸については、嗅細胞の嗅上皮における位置を反映してその転写量が決まる反発分子、Nrp2とSema3Fによって、発生期に時空間的な制御を受けている事が明らかになった。
神経地図形成に関してはこれまで、投射先に濃度勾配を示して分布する軸索ガイダンス分子と、軸索末端に発現する受容体の相互作用が重要な役割を果たすと考えられてきた。しかしながら当グループの一連の研究により、高等動物の嗅覚神経地図は、ターゲットには依存せずむしろ投射してくる嗅細胞軸索間の相互作用により、自律的に形成される事が明らかとなった。
