Research Abstract

H19年度は以下の2点についてそれぞれ成果を得た。以下に列記する。
DI RNAが樹状細胞に与える影響の解析:麻疹ウイルス株を用いた解析からワクチン株にDI RNAを含む株が多いこと、DI RNAが1型interferon(IFN)を強く誘導することを証明した(Shingai, Seya et al., J Immunol2007)。具体的にはEdmonston (ED)株はDI RNAを含みIFNを強く誘導したが、リバースジェネティクスにより人工的にDI RNAを含まないEdmonston株ウイルスを作製するとIFN誘導を殆ど起こさなかった。DI RNAのstem-loop構造にIFN誘導の鍵があると考え、この構造物をRNA polymerseで増やして如何なるIFN誘導経路を活性化するか検討した。結果はRIG-I/MDA5がstem-loop RNAのセシサーとして働くことが示された。配列特異的なsiRNA効果がstem-loopに認められるかを検討中である。
DI RNAの宿主認識機構の解析:麻疹ウイルスのDI RNAはRIG-I/MDA5をレセプターとしてIFN誘導を行うことが判明したが、これ以外の配列で他のRNA認識パターン認識レセプター(TLR3,TLR7,TLR8,RIG-I,MDA5,PKR,dicer-siRNA),を活性化しうるかを検討している。IPS-1(MAVS)KOマウスを用意し、CD46/CD150 TGマウス(はしかに罹るマウス)と交配してtriple mutantを作成する。CD46+/CD150+1MAVS-のマウスにED株(DIRNAを含む)を感染させてほかのレセプターが関与するかを解析する。MAVS経路以外のDI RNA認識経路が想定できればこれを確定する。現在マウスを用意できたところである。
