Research Abstract

ラットおよびマウスにおける宇宙飛行や後肢懸垂による後肢筋活動の抑制が脳における学習能や遺伝子およびタンパク質発現に及ぼす影響を追求した。3ケ月間国際宇宙ステーションで飼育したマウスおよび地上コントロールマウスの脳サンプルでは、遺伝子およびタンパク質発現を網羅的に分析した。地上コントロールに比べて、免疫・炎症反応や各種酵素活性等の代謝に関与する遺伝子発現が宇宙飛行群で高かった。逆に、異化作用や代謝に関係する遺伝子に低下したものも見られた。宇宙飛行に伴う発現増加は28種のタンパク質にも認められた。これらのタンパク質は、ミトコンドリア代謝、ATP合成および加水分解、カルシウム・カルモデュリン代謝、神経系、タンパク質・アミノ酸輸送に関係するものであった。ミトコンドリア代謝に関係するタンパク質には、発現量が逆に低下するものも認められた。このように遺伝子とタンパク質発現には、必ずしも一致しない反応もあったが、一般的にまず遺伝子発現が変化し、タンパク質合成に変化が起こる。したがって、タンパク質合成に適応が誘発されれば、遺伝子の役割は終わるわけであり、遺伝子には3ケ月の間に逆方向の発現が起きたものと推察される。地上での新生ラットの後肢懸垂は、迷路水槽を使って推測した学習能や神経新生が抑制されるという結果も得られた。長期間の宇宙飛行がマウスの脳機能にどのような影響を及ぼしたのかは不明であるが、抗重力筋活動が脳の機能に何らかの影響を及ぼすことは明らかとなった。このような知見はヒトにも応用できるはずであり、脳機能の維持亢進のたにも日常の身体運動が推奨されよう。
