Research Abstract

1)軟骨変性制御分子のクローニングと分子ネットワークの解明
治療標的分子の同定のために細胞外分子の検索を行ったところ、Notchシグナルが生理的および病的な軟骨内骨化を制御することを、時期特異的および組織特異的ノックアウトマウスの作成によって明らかにした。軟骨細胞におけるNotch/RbpjシグナルはHes1を介してMMP13を転写誘導することによって軟骨内骨化を制御していた。中でもHes1の関与は生理的軟骨内骨化よりもOAにおける病的軟骨内骨化に対して強いことから、副作用の少ない有望な治療標的分子であると考えられた。
2)重症度評価システムの開発
膝システムのKOACADが「膝関節診断支援方法及び装置並びにプログラム」として特許を取得した。このソフトは既に発売を開始し、先進医療を申請中である。また、股関節・腰椎X線自動定量評価システムのX線自動測定システムの開発が進んでおり、これらのシステムを用いてROADデータベースの解析を行なっている。
3)遺伝・環境因子の網羅的・系統的探索
変形性腰椎症の疾患感受性遺伝子の解明を、Affymetrix社製900k ArrayによるGWASを用いて行ない、数種類の候補遺伝子を同定した。引き続いて韓国、ヨーロッパのゲノム研究グループと共同研究によって、これらの候補遺伝子SNPのreplication解析を行っている。また、特に可能性の高い遺伝子についてはマウスゲノミクスを用いてその機能解析を行っている。
