マウスモデルを用いた消化器癌転移の研究

Research Abstract

1.我々はこれまでに、DNAマイクロアレイを用いて32個の転移関連候補遺伝子を同定している。本年度は、それらの遺伝子をそれぞれ安定発現させたColon26マウス大腸癌細胞株を樹立した。また、発現させた遺伝子が転移の初期過程である癌細胞の浸潤に関与するかを調べる目的でin vitroのinvasion assayを行なった結果、4遺伝子が浸潤を促進し、5遺伝子が逆に浸潤を抑制することが分かった。また、DNAマイクロアレイの結果をさらに詳細に検討したところNotchシグナルに関与する分子の発現が原発巣に比べて転移巣で変動していることが分かった。
2.Colon26細胞及びHCA7ヒト大腸癌細胞を使用し、Aes遺伝子の安定発現株を作成した。これらの細胞をマウス直腸に同所移植したところ、肝臓や肺への転移が有意に減少することが分かった。また、ルシフェラーゼレポーターを用いた解析により、AesがNotchシグナルを有意に抑制することを明らかにした。さらに、Colon26細胞を同所移植したマウスにNotchシグナルを遮断するγ-secretase阻害剤を投与すると転移が抑制されることが分かった。これらの結果からAes遺伝子はNotchシグナルを抑制することで大腸がんの転移を阻害することが明らかとなった。
3.リンパ節転移を最も容易に観察出来るB16F10細胞を用いて転移先周辺組織の変化を検討した。癌をドレナージするリンパ節(B16F10を接種したC57BL/6マウスの膝下リンパ節)においてはリンパ節辺縁洞が拡張すること、これら拡張した辺縁洞内へ癌細胞が流入してくる事を見出した。また、この周囲のT細胞領域では正常リンパ節と比べCXCL9,CXCL10の発現が亢進することが分かった。さらに、これらCXCR3リガンドはマクロファージやリンパ管内皮細胞に発現すること、原発巣におけるマクロファージはこれらのケモカインを発現せず、リンパ節特異的な発現上昇である事が示唆された。リンパ節辺縁洞に流入したCXCR3発現癌細胞は、発現上昇したケモカインと接触することにより、生存亢進、増殖のシグナルが入る事が考えられた。
