マウスモデルを用いた消化器癌転移の研究

Research Abstract

(1)前年度までに、大腸癌細胞株にAes遺伝子を強制発現させると、Notchシグナルを抑制することで転移能が阻害されることを明らかにした。より生理的なAesの役割を解析するため、siRNAを用いてAesをノックダウンした大腸癌細胞株をマウス直腸に移植したところ、肝転移巣の数が対照細胞株に比べて有意に増加していた。一方で原発巣の大きさは変わらなかったことから、Aesは癌細胞の増殖抑制以外の機序によって転移を抑制する、新規の転移抑制遺伝子であることが分かった。また、このマウス直腸接種モデルの原発巣において、宿主側血管内皮細胞がNotchリガンドを、癌細胞がNotchレセプターを発現していたことから、これらの細胞間でのNotchシグナルの伝達が転移に重要である可能性が示唆された。さらに、臨床標本においても、肝転移巣におけるAESの発現は同一患者の大腸癌原発巣よりも有意に低下していた(52症例中29症例、56%)ことから、ヒト大腸癌においてもAESが遠隔転移を抑制していることが示唆された。(2)B16F10メラノーマ細胞をfootpadへ移植したマウスにTAK-779(武田薬品)、AMG-487(Amgen)を投与したところ、膝下リンパ節への転移が抑制されること、これらの薬剤が転移巣における癌細胞の増殖を抑制していることが示された。また、癌細胞を移植したマウスのリンパ節を詳細に解析した結果、リンパ節辺縁洞が拡張していること、癌細胞はその領域内へ流入することを明らかにした。また、その周囲のT細胞領域では癌細胞の発現するCXCR3リガンドであるCXCL9,10の発現が亢進することを見出した。これらのCXCR3リガンドは原発巣には発現していないことから、リンパ節特異的に発現上昇したケモカインがリンパ節辺縁洞に流入したCXCR3発現癌細胞の生存、増殖を促進している可能性が示唆された。
