樹状細胞のウイルスパターン認識分子による細胞性免疫活性化機構

Research Abstract

ウイルスの産生する核酸パターンが樹状細胞(mDC)依存性にエフェクター(NK,CTL)を誘導する分子機構を解析し、以下の結果を得た。
1.NK活性化に必要な樹状細胞因子:マウス樹状細胞(mDC)-NK reciprocal activationの担当分子を同定した。この分子はpolyI:C刺激でmDCのTICAM-1(TRIF)経路依存性に発現する膜分子で、IRF-3の活性化が必須であった。従って、IRF-3-inducible NK activating molecule(ONAM)と名付けた(J Exp Med 2010, in press)。INAMは樹状細胞による抗がんNKの活性化の起動因子であった。INAMはTICAM-1経路以外にIPS-1細胞内経路によっても誘導された。mDC-NK reciprocal activationの担当分子でもあり、種々の微生物成分で樹状細胞がNK活性化を誘導する責任分子でもあった。
2.CTL誘導に必要な樹状細胞因子群:mDCではTLRアダプター,MyD88,TICAM-1,RIG-IアダプターIPS-1がCTLの誘導に関与する。各KOマウスのmDCとCFSEラベルしたOT-1,polyI:C刺激した脾細胞を用いてcross-primingを査定した。結果、TICAM-1が主要なcross-priming誘導アダプターと判明した。この応答にはIRF-3/7両方が関与した。この結果はウイルス感染が樹状細胞以外の細胞に起きた時、CTL誘導が効率よく起動することを示唆する。
