細胞内ナノドメイン機能制御による心不全治療法の開発

Research Abstract

不全心筋細胞において、細胞内ナノドメイン機能制御を行うための分子標的を同定した。プロテオーム解析用に精製された心筋細胞筋小胞体(SR)をinuiらの方法に基づいた段階的遠心法+密度勾配超遠心法でlongitudinal SRドメイン、Junctional SRドメインを分離精製し、構成蛋白をショットガンプロテオミクスで同定した。筋小胞体構成蛋白約200を同定した。もっとも高頻度に検出された蛋白は、SERCA2a、 GRP78、 GRP94、 sarcalumenin、 carsequestrin, phospholamban(PLN)などであった。同定された蛋白の大部分はSRないしendoplasmic reticulum(ER)に豊富に局在することが報告された蛋白で、機能未知の蛋白も30個程度認められた。
また従来から解析を行っている蛋白ホスファターゼ1については、PP1βアイソフォームがもっとも筋小胞体分画に多いことを同定した。アデノウイルスを用いたRNAiにより、PP1βをノックダウンすると心機能が改善した心筋細胞のPP1アイソフォームをノックダウンするとPP1βがもっともPLNのリン酸化状態を変化させた。PP1βは各種不全心で発現量が増加しており治療標的になる可能性があると考えられた。
また、心不全においてはリアノジン受容体が不安定となり、細胞内のCa2+循環が不整となるが、このRyRを安定化させる化合物JTV519のアミノ酸結合部位を同定した。同部位のアミノ酸配列を筋小胞体と反応させてもRyRが安定化することより、分子標的治療として有効であると考えられた。
