| Research Abstract |
本研究の目的が、セメント質誘導性機能性ペプチドの開発であるが、平成20年度には、共同研究者の芝らが開発した研究により、細胞のインテグリンなどと結合するRGDモチーフのタンパク質とチタンに結合するタンパク(TBP-1)を応用し、チタン上に両モチーフを含む合成タンパク質をMolclaftにより作製し、in vitroによるデータを重ね発表し、Kokubun, K., Kashiwagi, K., Yoshinari, M., Inoue, T., Shiba, K. Motif-programmedarttificaila extracellular matrix, Biomacromolecules, Volume9, Number11, pages3098-3105,2008に掲載された。この結果、従来タンパク質がつくことの無かったチタンインプラントの表面にTBP-1を介して様々なタンパク質をつけることが可能となった。つまり、これらのことは、上皮性の強固な結合を可能にし、外部からの細菌侵入にたいて、リスクを減らすこととなり、加えて、骨誘導タンパクを応用することができれば、骨粗鬆症や骨量不足の患者にも、インプラントを応用することが可能となる。しかし、課題も残り、創傷の治癒を考えたとき、出血・凝固、白血球による創内浄化、肉芽組織による組織再生、そして再構築の適切な時期に、必要なタンパク質が徐放され、細胞に働く必要がある。そのためには、Drug Delivery systemの構築が要求され、適切な時期に適切なタンパクがインプラント周囲の細胞に働く必要がある。また、本当にこれらチタンに結合したタンパク質が生体内においてその機能を発揮するか否かを早急に検討する必要がある。そのために、次年度には動物実験を行い、検証していく必要がある。先ず、従来より良く知られているBMPなどを使い、実証し、その後タンパク質誘導タンパクの検索と、その応用を同様に行っていく必要がある。
