Research Abstract

本研究は,植物の光形態形成を制御している光受容色素蛋白質フィトクロムの発色団である開環状テトラピロール(ビリン系化合物)の合成を基盤として,フィトクロムの構造と機能を解明するものであり,最終的には農産物の増産などに寄与しうるものと考えられる。具体的な研究計画としては
(1)フィトクロム発色団のAB環及びCD環部位の立体化学を固定したビリベルジン(BV)誘導体を合成し,再構成実験によりPr型及びPfr型フィトクロムの立体化学の解明を目指す。
(2)フィトクロム発色団のCD環部位を固定したBV型発色団を用いるPr型及びPfr型フィトクロムの結晶化とX線結晶構造解析を目指す。
(3)AB環及びCD環部位の立体化学を固定したフィコシアノビリン(PCB)誘導体の合成と試験管内及び発色団欠損変異株を用いる生体内再構成並びに表現型(phenotype)の解明を目指す。
(4)植物フィトクロムの簡便精製法の実現を目指して,ビリン系発色団担持型アフィニティー・クロマトグラフィーを確立する。
(5)光学活性なPCB発色団の新規一般合成法の確立を目指す。
