Research Abstract

本研究課題は、ES-体細胞融合細胞からES細胞由来の染色体のみを取り除くことで体細胞由来の多能性幹細胞を作り出すための基盤技術の開発を目的としている。方法として、(1)染色体除去の新分子ツール、染色体除去カセット(CEC)の開発、(2)染色体除去の高効率化をもたらす技術の開発、(3)ECE-ES細胞ライブラリーの作製、(4)ES細胞全染色体へのCECの導入、(5)ホストES細胞の開発、(6)ヒトES細胞と体細胞の新細胞融合技術の開発、を挙げていた。マウス細胞を用いた研究成果として、(1)ECE-ES細胞ライブラリーの作製の成功、(2)ES細胞へ40コピー以上のCECの挿入、(3)染色体の一斉除去による細胞死の誘導、(4)染色体除去技術を用いた遺伝子量バランスの重要性、を明らかにすると共に、(5)ES細胞:体細胞=1:1の融合システムの開発を行った。
