Research Abstract

蛍光相関分光法(Fluorescence Correlation Spectroscopy; FCS)を用いた転写因子活性の確立と関節リウマチ、アレルギー疾患への応用。
1,蛍光相関分光法(Fluorescence Correlation Spectroscopy; FCS)を用いた測定系の確立のため転写因子活性の測定系の基礎的検討を行った。主にリコンビナント蛋白を用いて行い良好な標準曲線を得た。
2,CSによる健常人培養リンパ球核抽出液を用いた転写因子活性測定系の確立。さまざまな条件化で正常人リンパ球を活性化し、核抽出物を単離し転写因子活性を測定した。同時に市販の転写因子ELISAを用いてその確からしさを検討した。
3,FCSによる健常人及び患者末梢血単核球の転写因子活性測定。その予備段階として、関節リウマチ患者末梢血から単核球を採取し、核内蛋白を抽出し、さまざまな転写因子をELISA系で測定した。同時に健常人をコントロールとした。測定項目は、NFkB、NFAT、AP-1、STAT1、STAT3、IRF-1である。なかでもRA患者においてはNFkB、STAT3の高値例が存在することが明らかとなった。そこで、さまざまな治療群における転写因子活性を測定する計画とし検体を採取している。
4,来年度は未治療RA群の検討、生物製剤治療前後の検討、生物製剤使用郡における反応性との関連に焦点を当てて検討していく予定である。
