腎臓における低酸素応答経路の網羅的機能解析とその治療応用

Research Abstract

VEGF の native promoter の下流に luciferase をつないだレポーターベクターを用いてiRNA libraryをスクリーニングすることによって見出した新規VEGF調節因子 MAP3K6は、stable cell line の作成などにより特異的にVEGF を調節することが判明した。また、培養血管内皮細胞を用いてMatrigel にて capillary formation をみるアッセイを行い、MAP3K6による VEGF の発現調節が、生理的に役割を果たしていることを示した。更に、MAP3K6をノックダウンしたHeLa細胞をヌードマウスに移植して解析し、MAP3K6によるVEGFの発現調節が tumor genesisに関与していることを証明した。
これと並行して、慢性低酸素腎の GeneChip による網羅的遺伝子発現解析を行った結果見出したmetallothionein に関する解析も進めた。metallothioneinはそれ自身がhypoxia-inducible factor (HIF) を活性化する新しい機能を持つことが、添加実験および高発現実験によって示された。更に、matallothionein による HIF の活性化は ERK/mTOR pathway を介しており、Akt とは関係ないことが分かった。また、同様に網羅的遺伝子発現解析により、低酸素腎において腎臓に内因性にヘモグロビンが発現しており抗酸化能を持っていること、一方 Cu/Zn SOD の発現は減少しておりこれがTNF-alpha によることも見出した。
