Research Abstract

平成20年度はスキャフォールドを用いない間葉系幹細胞由来三次元人工組織(Tissue engineered construct; TEC)の骨・軟骨分化誘導促進刺激をIn vitroおよびln vivoにて検証した
骨分化に関しては、カルシウムイオン溶液とリン酸イオン溶液への交互浸漬法によるハイドロキシアパタイト(HaP)結晶沈着技術がTECの骨分化誘導を再現性高く誘導することを平成19年度の研究で見出したが、20年度では、このHap結晶沈着TECを成熟家兎軟骨下骨欠損部に移植し、コントロール(Hap非沈着TEC)に比して有意な骨新生の亢進効果を確認した。
本成果はHap結晶沈着TECの骨再生バイオマテリアルとしての有用性を示唆するものである。
また軟骨分化に関しては骨形成因子(BMP-2)による刺激によりTECの軟骨分化が促進される効果の至適条件の検討をBMPの濃度および刺激期間の観点より行った。TECはBMP-2に対し濃度依存性に軟骨分化を亢進させるが100ng/mlでその効果はプラトーとなった。また刺激期間については14日までは期間依存性にTECの軟骨分化は更新したが、それ以上の期間ではその効果はプラトーであった。また組織学的には培養期間によらず中心壊死は存在しなかった。本成果によりTECを作成する際のBMP-2による前処理における至適条件が示唆された。
