ラット脊髄後角の痛み伝逹制御におけるアラキドン酸カスケードの役割

Research Abstract

末梢から後根を通って脊髄に入力する痛み伝達の制御は、脊髄後角第II層(膠様質)のシナプス伝達修飾によりなされることはよく知られている。ホスホリパーゼA_2(PLA_2)活性化ペプチドであるメリチンを用い、PLA_2活性化はアラキドン酸カスケードを介して膠様質の興奮性や抑制性のシナプス伝達を促進することを明らかにしている。この仕組みの詳細を知るために、成熟雄性ラットから作製した脊髄横断スライス標本の膠様質ニューロンにホールセル・パッチクランプ法を適用して、様々な薬物の作用下でメリチンがGABAやグリシンを介する抑制性シナプス伝達に及ぼす作用を調べた。Na^+チャネル阻害剤(テトロドトキシ)、グルタミン酸受容体阻害剤(CNQXとAPV)、細胞外の無Ca^<2+>、α_1アドレナリン受容体阻害剤(WB-4101)、マスカリン性とニコチン性のアセチルコリン受容体阻害剤(それぞれアトロピンとメカミラミン)の各々は、メリチンによるGABA作動性シナプス伝達の促進を抑制した。これらの阻害剤の存在下や無Ca^<2+>液中でメリチンによるグリシン作動性シナプス伝達の促進は影響を受けなかった。以上より、メリチンはグリシン作動性のシナプス伝達を直接促進するが、メリチンによるGABA作動性シナプス伝達の促進には、α_1アドレナリン受容体、マスカリン受容体およびニコチン受容体の活性化が関与していることが示された。PLA_2活性化によるグリシン作動性シナプス伝達の促進にはリポキシゲナーゼ代謝物の関与が明らかにされているが、この促進は、細胞外の無Ca^<2+>ばかりでなくCa^<2+>チャネル阻害剤(La^<3+>)、さらにCa^<2+>誘起Ca^<2+>放出機構の阻害剤(ダントロレン)やIP_3誘起Ca^<2+>放出機構の阻害剤(2-APB)により有意に抑制されなかった。これよりグリシン作動性シナプス伝達の促進作用はCa^<2+>非依存性であることが示唆された。
