| Research Abstract |
個細胞を固定化して細胞の機能診断を行ない,温度刺激によって細胞を非侵襲的に選択的に剥離・回収する新規な細胞診断デバイスを開発した。ガラス基板上のサイズやパターンを制御した金スポットに温度刺激に可逆的に応答するpoly(2-hydroxypropylmeth-acrylamide-co-methylmethacrylate),ガラス面には細胞非接着面としてのpoly[2-(methacryl-oyloxyjethyl phosphoryl choline]をそれぞれ修飾した。細胞播種により個細胞をアレイ表面に固定化できることを初めて見出した。温度刺激で90%以上の接着細胞が非侵襲的に剥離した。DNAトランスフェクションを90%以上の高効率で行える等,本細胞マイクロアレイが診断と分取用デバイスとして有用でかつ実用化可能であることを示した。
