Research Abstract

ウシ副腎髄質由来のクロマフィン細胞(1,2)とマウス骨芽細胞(3)の実験を試みた。
1.クロマフィン細胞を低張溶液にインキュベートすると、その直後にその細胞容積は一過性の増加を引き起こし、その後次第に液交換前の容積に回復した。1.5Tの変動磁界曝露およびサイトカラシンDを添加すると、F-actin量が減少したが、低張溶液への置換は交換直後の容積増加の最大値の増加と、回復時間の遅延が認められた。この影響は複数のF-actin重合阻止剤添加でも確認された。このことは磁界がactinの重合、脱重合に影響を及ぼすことを示唆する。
2.高周波電磁界曝露がアセチルコリン添加および高K^+溶液置換による細胞内Ca^<2+>濃度増加に及ぼす影響を調べた。細胞内Ca^<2+>は両者において一過性の増加を示し、時間経過と共に次第に減少する傾向を示した。このとき周波数は438.5MHzに一定とし、電磁場強度を26.6から54.6Wに変化させた。行ったすべての実験で、高周波電磁場曝露は上記の細胞内Ca^<2+>増加には有意の影響をおよぼさなかった。これらの結果は438.5MHzの高周波電磁場は小胞体からのCa^<2+>放出および細胞膜のCa^<2+>チャネルを介するCa^<2+> influxには影響をおよぼさないことを示した。
3、マウス骨芽様MC3T3-E1細胞の分化における極低周波磁界(ELF-EMF : 3mT, 60Hz)の影響を調べた。ELF-EMF曝露によりコラーゲン合成が刺激されるが、これはp38MAPK経路の介入により引き起こされる。またPI3K経路はこのELF-EMF曝露により誘導されるコラーゲン合成の抑制に関与し、このPI3K経路の抑制はコラーゲン合成を促進する可能性があり、磁界の関与を示唆した。
