Research Abstract

オピオイド化合物が受容体に結合する際、塩基性窒素が必須であることは既に知られているが、今回、窒素の非共有電子対の配向性が特に重要であることが明らかとなった。すなわち、モルヒネと同様のアキシアル方向にあると結合するが、それとは異なった方向(エクアトリアル方向)にあると結合しないか、結合しても非常に弱いことが明らかとなった。また、作動薬および拮抗薬の溶液中における窒素置換基の配向性を検討したところ、作動薬と拮抗薬とで窒素置換基が空間的に占める位置が異なることが示唆された。
