薬物トランスポータの膜局在決定因子の同定と極性輸送機構の解明

Research Abstract

小腸や腎臓などの上皮細胞では、血管側側底膜と管腔側刷子縁膜に機能特性の異なる薬物トランスポータが局在し、薬物の臓器移行性や薬物輸送の方向性(吸収・分泌)の決定に重要な役割を果たしているが、その細胞膜局在を決定する分子機構については未だ不明な点が多い。本研究では有機カチオントランスポータ(MATE1,0CT2)をモデル蛋白質として選び、それぞれの膜局在を規定する因子(薬物トランスポータ結合蛋白質)を探索、同定することを試みる。まず、有機カチオン輸送活性を有しない培養腎上皮細胞MDCKにMATE1,0CT2を共発現させたところ、OCT2は側底膜、MATE1は頂側膜に局在し、側底膜側から頂側膜側への分泌方向に対応する方向選択的な有機カチオン輸送が認められた。そこで、MATE1のN末端またはC末端側にgreen fluorescent proteinまたはV5をtagとして付加したコンストラクトを作成し、MDCK細胞に安定発現させたところ、N末端にtagを付加した場合には頂側膜に発現するのに対し、C末端にtagを付加したものではほとんど発現が認められないことが明らかになった。従って、MATE1のC末端側の領域がその頂側膜への発現に重要であることが示唆された。また、頂側膜に発現するMATE1は推定される分子量よりも小さく、細胞膜移行の過程で何らかのプロセシングを受けているものと考えられた。現在、MATE1の結合蛋白質の同定を進めており、さらにMATE1の生合成後の細胞内輸送経路、細胞膜発現挙動について明らかにしていく予定である。
