薬物トランスポータの膜局在決定因子の同定と極性輸送機構の解明

Research Abstract

小腸や腎臓などの上皮細胞では、血管側側底膜と管腔側刷子縁膜に機能特性の異なる薬物トランスポータが局在し、薬物の臓器移行性や薬物輸送の方向性(吸収・分泌)の決定に重要な役割を果たしているが、その細胞膜局在を決定する分子機構については未だ不明な点が多い。本研究では有機カチオントランスポータ(MATE1,OCT2)をモデル蛋白質として選び、検討を行った。まず、有機カチオン輸送活性を有しない培養腎上皮細胞MDCKにMATE1,OCT2を共発現させたところ、OCT2は側底膜、MATE1は頂側膜に局在し、側底膜側から頂側膜側への分泌方向に対応する方向選択的な有機カチオン輸送が認められた。そこで、MATE1のN末端またはC末端側にgreen fluorescent proteinまたはV5をtagとして付加したコンストラクトを作成し、MDCK細胞に安定発現させたところ、N未端にtagを付加した場合には頂側膜に発現するのに対し、C末端にtagを付加したものではほとんど発現が認められないことが明らかになった。従って、MATE1のC末端側の領域がその頂側膜への発現に重要であることが示唆された。そこで、MATE1のC末端側の欠失変異体を構築し、細胞に発現させて局在を観察した結果、MATE1の細胞膜局在に重要と考えられるC末端側領域を決定することができた。現在、この領域に特異的に結合する蛋白質の同定を進めている。
