Research Abstract

上皮細胞間をシールするタイト結合は、細胞間隙を通る分子やイオンの選択的透過性を制御するバリア機能と、細胞極性を規定するフェンス機能を担い、その破綻は炎症性腸疾患やがん等様々な病態の原因や修飾因子になると考えられている。特にclaudin(CLDN)は、タイト結合の形成に必須の蛋白で、CLDNsは20種類以上からなる遺伝子ファミリーを構成している。CLDNは、第一細胞外ドメインの荷電アミノ酸の位置や数によって異なるイオン透過性を有することが示唆されている。そこで今回我々は、「細胞間隙における分子通過、とくにカルシウム吸収を制御するメカニズム」の解明に結びつけることを目的として、正常マウスおよびビタミンD受容体ノックアウト(VDR KO)マウスを用いて、十二指腸、空腸、回腸、結腸におけるCLDNsの発現パターンを検討し、さらにビタミンD依存性にカルシウムの透過性を亢進するヒト腸上皮細胞株Caco-2を用いて、それぞれのCLDsがカルシウム透過性に及ぼす影響を検討した。
1)マウス腸管においてCLDN7,8,12,13,15が発現することを見出した。
2)これらのCLDNsは、その発現部位と発現量がそれぞれ異なり、特異的発現パターンと細胞内局在を示した。
3)カルシウムイオンが吸収される小腸では、CLDN12の発現が高いことが明らかになった。
4)ビタミンD受容体ノックアウト(VDR KO)マウスの腸組織におけるCLDNの発現を解析したところ、CLDN2,12の発現が著明に減弱していた。
5)ヒト腸上皮細胞株Caco-2をビタミンDで処置するとカルシウムイオンの透過性が亢進し、CLDN2,12の発現が誘導された。さらにRNAi法や強制発現法を用いて、CLDN2,12がカルシウムイオンのparacellular channelとして働くことを確認した。
以上から、ビタミンDがVDRを介してCLDN2,12の発現を誘導して、小腸におけるカルシウムイオンの細胞間隙通過を制御すると考えられた。
