| Research Abstract |
癌の増殖の本体は「癌幹細胞」にある。「癌幹細胞」は「幹細胞/前駆細胞」に由来するのか?本研究では、肝臓の「肝幹細胞/前駆細胞」とされるoval cellの関与するラット肝発癌モデルを用い、早期の前癌性病変が「肝幹細胞/前駆細胞」に由来し得るか検索した。その結果(1)oval cellに発現する幹細胞マーカーThy-1, EpCAM, c-kit, CD34, CD133の各発現細胞がoval cellの増生から前癌性病変の発生に至る過程に関与する事を免疫組織学的に示した。(2)全身緑色蛍光を発するラットに肝発癌処置を行い増生したoval cellを含む細胞から磁気細胞分離法にて分離したThy-1, EpCAMの各発現細胞を無蛍光の野生型ラット肝に移植すると緑色蛍光を持つ前癌性病変が発生した。これはThy-1またはEpCAM発現「肝幹細胞/前駆細胞」が肝前癌性病変を発生し得る事を示す最初の報告である。
