ベトナムの枯葉剤濃厚散布地区におけるダイオキシン汚染の細胞遺伝学的研究

Research Abstract

金沢大学:城戸照彦教授は2002年より、社会経済状況が類似している2つの地域、つまり、北緯17度線をはさんで最汚染地区(Quang Tri省Cam Chinh地区:旧南ベトナム領)と非汚染=対照地区(Ha Tinh省Cam Phuc地区:旧北ベトナム領)を選定し、両地区の住民それぞれ4100人前後を対象とした大規模な疫学調査(死産率・先天異常発生率・男女の出生比率・栄養状態)と環境調査(土壌・水質・低質・魚家禽豚のダイオキシン濃度測定)を行った。更に、この中の150-190人については生体試料(血液・脂肪組織・母乳)を採取して、ダイオキシン類の濃度分析を行った、その結果、疫学調査、環境調査の何れにおいても、汚染地区と非汚染地区では有意差があり、生体試料検査では汚染地区住民は非汚染地区住民に比べ、3-4倍高いダイオキシン濃度を示していることが判明した。私たちは共同研究として、既に母乳中ダイオキシン類濃度の測定が行なわれている両地区の母親から末梢血を採血し(汚染地区:50人、対照地区:19人)、姉妹染色体分体交換(sister chromatic exchange:SCE)指数(現在、最も鋭敏なDNA毒性検査とされている)を調査した。その結果、(1)汚染地区(45人)と対照地区(19人)ではSCE指数に統計学的に明確な差が認められる(2.34 vs 1.53)、(2)汚染地区内のダイオキシン類高濃度群と中程度濃度群の間で、SCE指数に統計学的に有意差が認めらる(2.50 vs 2.19)、(3)SCE指数とダイオキシン類濃度との間には、log値を取った場合に緩やかな相関が認められる(logSCE=0.1713xlogTEQ+0.156,r=0.463)、ことを明らかにした。本年はダイオキシン異性体のデータを金沢大から頂き、SCE指数との関連について、多変量解析を用いて分訴する予定であった。しかし、ダイオキシン異性体のデータが届いたのはH21.6.3であった。現在、その解析を行っている。この結果は論文化する予定である。
