心血管疾患におけるSDF-1/CXCR4システムの役割の解明と治療への応用

Research Abstract

我々は、自己骨髄細胞を用いて重症下肢虚血や心筋虚血患者を対象に血管再生療法を施行している。そこで、治療の有効性を高め、かつ患者への侵襲の少ない新たな心血管再生療法の確立を目的として、SDF-1/CXCR4システムに着目して研究を行った。培養により骨髄細胞のCXCR4発現が有意に増加することを見出し、この発現増加作用には、マウス血清に骨髄細胞が暴露されなくなることが、その機序の一部として関与していることを明らかとした。このCXCR4発現の増強された培養骨髄細胞は新鮮骨髄細胞と比較してマウス下肢虚血モデルにおける血流改善並びに毛細血管形成を有意に促進することが示された。この血流改善機序として、虚血部位へと移植した骨髄細胞がそこで発現しているSDF-1(CXCR4リガンド)と相互作用してより局所に留まりやすくなることと、CXCR4高発現の骨髄細胞がIL-1βやVEGFなどの血管新生因子をより多く産生することを明らかとした。今後、SDF-1/CXCR4システムの役割と血管新生における詳細な機序が解明されることにより、これを標的としたより効果の高い血管新生療法が開発されることが期待される。
