腎臓における小胞体ストレスの病態生理学的意義の解明

Research Abstract

申請者らはこれまでに、腎臓特異的セリンプロテアーゼインヒビター"メグシン"の高発現ラットが腎臓細胞の小胞体ストレスに陥って腎不全を発症する「新規腎不全モデルラット」であることを明らかにすると共に、腎臓において小胞体ストレスが腎病変形成に深く関与する可能性を示唆してきた。そこで本研究では、メグシン高発現ラット、いわゆる腎小胞体ストレスモデルラットの腎臓を指標にして、各種腎疾患モデル動物における小胞体ストレスの動態を分子レベルで比較検討し、小胞体ストレスの腎疾患における病因論解明を試みた。
その結果、ネフローゼモデル(puromycin腎症ラット)や糸球体障害モデル(Thy-1腎炎ラット)において小胞体ストレスのマーカーとも言える小胞体ストレス応答性シャペロン分子(ORP150,GRP78)の発現が、puromycin腎症ラットでは尿細管に、Thy-1腎炎ラットでは糸球体に誘導されることを見いだした。さらに急性腎不全モデルである虚血再潅流ラットにおいても同様に、病変部の尿細管においてORP150,GRP78の発現亢進が認められた。これらのことから、尿細管においても、糸球体においても障害時に小胞体ストレスを伴う可能性が示された。そこで、Thy-1腎炎ラットにおける小胞体ストレスの経時的変化を詳細に検討したところ、1)小胞体ストレス応答性シャペロン分子の発現は糸球体障害の程度に伴って過剰に亢進すること、2)小胞体ストレス回避機構の一つである蛋白翻訳抑制因子(eIF2α,PERK)が著明に活性化(リン酸化)すること、3)小胞体ストレスの過剰変動をER stress preconditioning(Thy-1腎炎誘導前に腎臓に影響のない軽度の小胞体ストレスを誘導しておく)によって軽減させることで、Thy-1腎炎の病像を改善できることなどを明らかにし、腎病変部における小胞体ストレスの病態生理学的関与を示した。
