| Research Abstract |
細胞外マトリックスの1つである。パールカンの遺伝子欠損によりSchwartz-Jampe症候群(SJS)を発症する。我々は本症候群における骨格筋の持続収縮、ミオトニア症状の発症機構についてモデルマウスを使って解明している。その過程でいくつかの複合要因が考えられ、その一つが筋の易収縮性である。また、本症候群に特徴的な眼論筋や口輪筋の持続収縮、筋の肥大、type1 fiber predominancy等の事象に関連して細胞内カルシウムの変動が重要な意味を持つと考えた。パールカン欠損と野生型マウス、及びその初代培養筋細胞の実験を行い、1) in vivoにおける筋線維タイプの存在比率や、筋横断面積の比較、2) in vitroにおける細胞内カルシウム濃度変動の差異からパールカンがカルシウム維持機構に何らかの関与をしている可能性が示唆された。
