| Research Abstract |
本研究では2型糖尿病での膵γ細胞におけるインスリン分泌の相対的低下の発症に着目した。今回我々は、フォークヘッド転写因子FoxO1に焦点を絞り、その活性型および抑制型変異体を膵β細胞で特異的に過剰発現させるマウスを作製しその表現型を解析するとともに、生体内での膵β細胞での遺伝子変化に与える影響を検討した。膵β細胞特異的に活性型FoxO1を過剰発現させると空腹時の高血糖を認めた。また、抑制型FoxO1をLepr^(db/db)マウスのβ細胞において過剰発現させると耐糖能の改善が認められた。γ細胞においてはp27遺伝子の発現には影響は認められなかった。
