Research Abstract

(1)ATL細胞におけるDNA修復阻害因子hnRNP B1の過剰発現とその意義
肺がんの早期から過剰発現することを見出したhnRNP B1タンパク質は、細胞内でDNA依存性キナーゼ複合体に結合することを見出している。hnRNP B1はATLの70%以上の症例において過剰発現していた。その発現は慢性期から増加し、急性期、リンパ腫型においては約8割の症例において発現亢進することが明らかになった。ATL細胞を用いた解析ではhnRNP B1はDNA依存性キナーゼに結合しその酵素活性を阻害すること、hnRNP B1の量と非相同末端修復(NHEJ)の活性は逆相関の傾向にあることを明らかにした。
(2)新規抗がん剤SMS-06によるATL細胞に対する抗腫瘍効果の解析
SMS-06はIC_<50>として50-100nMの濃度で種々のATL細胞の増殖を抑制した。その機序としてG2/M停止とその後のアポトーシスの誘導によることを明らかにした。またSMS-06はトポイソメラーゼIIの阻害作用に加えてhnRNP B1の存在下において放射線照射によるDNA修復を阻害する作用を持つことを新たに見出した。さらにSCIDマウスへのATL細胞の移植系において腫瘍の増殖を抑制作用を確認した。複数の分子標的を有するSMS-06はATLの治療薬剤開発のためのリード化合物として臨床応用への検討を進めている。
