| Research Abstract |
生殖幹細胞に特異的に発現する遺伝子のうち7分子(Fth1,Cryab, Spp1, Bcap31, Arhgap1, Ctla2a, Serpina3g)が造血幹細胞にも特異的に発現していることを定量PCR法により明らかにした。またこれらの分子について、組織学的解析を行った結果、毛根部の幹細胞領域と思われるバルジ部位に特異的に発現している遺伝子として、Spp1を同定した(Takuo Mizukami et al, Stem cell & Dev.,17:67-80, 2008)。このことは、多くの体性幹細胞において、幹細胞機能に深く関与していることが示唆された。同定されたSpp1遺伝子の機能を解析するため、Spp1遺伝子欠損マウスを入手し、血液学的解析を行った。これまでのところ、Spp1欠損マウスにおいて、造血幹細胞の激的な増加が、ニッチと考えられている骨内膜とは異なる部位で出生後4週まで続くことが観察している。Spp-1がニッチにおいて未分化性維持を負に制御しているのではないかと考えられる。
さらに、生殖幹細胞に発現するHE4(Homo sapiens epididymis-specific)がマウス造血幹細胞(CD34陰性KSL細胞)に特異的に発現しており、発現量はβ-actinとの発現量の0.01倍であり、Tal1(造血幹細胞特異的転写因子)の1000倍の発現量であることを明らかにした。またヒトCD34陽性CD38陰性分画でも特異的に発現しており、β-actinとの発現量の0.003倍であった。これまでにHE4にはWAP(whey scidic protein)domainが含まれており、他の類似分子の機能から、プロテインインヒビターであることが想定されるが、幹細胞特異的分子としての機能に関与していることも想定される。トランスジェニックマウスの作出を行い、血液学的機能解析を継続して行っている。
