Research Abstract

PI3K/AKTシグナル伝達経路は成長因子のリン酸化シグナルを細胞内に伝達し、細胞の増殖や浸潤、抗アポトーシス作用に関わっている。このシグナル伝達を抑制するフォスファターゼであるPTENは前立腺癌、子宮内膜癌、神経膠芽腫、消化器癌など、様々な癌で発現の異常が報告されている。またPTENの胚細胞での変異はCowden病やBannayan-Zonana症候群などの遺伝病を生じ、甲状腺がんや乳癌の頻度が著しく上昇することが知られている。我々は胃癌におけるPTENやAKT/PI3Kのシグナル経路の異常を系統的に解析し、新しい分子標的としての意義を検討している。その過程で、この経路の活性化が低分化胃癌の治療に重要な意義をもつ可能性を見いだした。
