一酸化窒素の非シナプス型神経伝達に注目した麻酔薬の作用機序の解明

Research Abstract

ラットの線条体に於いて、in vivoマイクロダイアリシス法による脳内一酸化窒素(NO)代謝産物測定の技術を確立し、0.1pmol 1-1程度の検出感度でNOの変化を高精度で追及できるようになった。
また、無麻酔自由行動下の実験モデル動物に対して、臨床において一般に用いられるプロポフォール、セボフルランの細胞外NO濃度に及ぼす影響に関する研究を開始し、プロポフォールがNOを減少させる一方、NOドナーであるニトログリセリン、ニトロプスシド投与によるNO放出を、より増加させる作用のある事を発見した。この作用は、セボフルランによる麻酔では観察されず、麻酔薬の特性による差異を検討する因子として注目に値すると考えられる。今年度は、特に量依存性の証明に関して、実験を進めてきた。これらの依存性も確認でき、22年度からは新しいテーマに挑戦する。
以上の所見は未だ現象論の域を出ないが、本研究に於いて初めて明らかにされた事象であり、麻酔薬の作用機序解明の手掛かりになると考えられる。また、各種の薬剤の脳保護作用、神経毒性作用の解明にも役立つであろう。
成果の一部は既に国際学会へも発表しており、既に論文発表の準備をおおよそ終了している。22年度中には最初の成果が国際的学術誌に掲載されると予想する。
