Research Abstract

We transfected expression vector containing IL6 mRNA gene into human androgen dependent prostate cancer cell line (LNCaP). The secretion of IL6 into supernatant in cultured medium from IL6 gene transduced LNCaP (LNCaP/IL6) was confirmed by enzyme linked immuno-solvent assay (ELISA). At the same time, only expression vector was transfected into LNCaP as control (LNCaP/Co).
There was no significant difference in in-vitro growth among LNCaP, LNCaP/Co and LNCaP/IL6. However, when they were inoculated to nude mice, LNCaP/IL6 showed significantly rapid tumor growth compared to LNCaP and LNCaP/Co. Since LNCaP is androgen dependent cell line, tumor retardation was observed after castration in nude mice. Interestingly, LNCaP/IL6 showed more severe retardation by castration compared to LNCaP and LNCaP/Co.
