Research Abstract

OPG遺伝子欠損マウス(14週齢雄)に対して、HX531を混和(0.03%)した飼料を1匹当たり毎日3g、30日間与える実験を行った。その結果、HX531の投与は大腿骨皮質骨部における著明な骨量増加作用が認められた。この効果は、我々が、既に報告したビスフォスフォネート(骨吸収阻害薬)の皮下注射実験と同様の強いものであった。一方、RXRアゴニスト作用が報告されているHX630とPA024の投与においても、骨量増加作用が認められた。以上の結果から、新規レチノイドX受容体(RXR)アンタゴニスト・アゴニスト共に、in vivoにおける破骨細胞による骨吸収を阻害することにより、強い骨量増加作用を示すことが明らかとなった。
