Research Abstract

大規模化合物ライブラリーから何らかの機能変化(フェノタイプ)を誘導できる低分子化合物を同定し、その化合物の標的分子の同定を通して生命現象を解明しようというケミカルバイオロジーという手法が確立しつつある。免疫学研究においてケミカルバイオロジー的な手法が有用である可能性については、免疫抑制剤シクロスポリンの単離により、その標的分子シクロフィリンが同定されたことなどからも明らかである。そこで、Bリンパ球およびマクロファージの機能を制御する化合物の単離を試みた。Bリンパ球については、B細胞株WEHI-231や正常B細胞を用いて、その活性化や細胞死を制御する化合物のスクリーニング系を樹立した。また、マクロファージについては、サイトカイン産生を制御するスクリーニング系を樹立した。さらに、CD22はB細胞の免疫応答を制御する重要な分子であるが、α2.6シアル酸を認識するレクチン活性を持つ。そこで、α2.6シアル酸産生に必須の糖鎖転移酵素ST6GalIの発現を欠損する細胞株J558Lと、ST6GalI遺伝子を導入しα2.6シアル酸の産生を回復したJ558LST6細胞を用いて、フローサイトメトリーにより高感度にCD22とα2.6シアル酸の反応を阻害する化合物を同定するスクリーニング系を樹立した。ついで、化合物ライブラリーのスクリーニングを行ない、CD22とその糖鎖ブガンドの反応を阻害する化合物を同定するっとともに、種々のシアル酸誘導体とCD22の反応性を解析し、α2.6シアル酸に比べ数百倍高い親和性でCD22に結合する化合物の単離に成功した。
